One donor.
Many doses.
Off-the-shelf, ready
for patients
Sofi-cel, Allotera’s allogeneic, gene-edited CAR-T therapy, is built from a single healthy donor and put on the shelf, ready to fight T-cell cancers.
The Off-the-Shelf Journey · 5 Steps
Step 01 / Source
HEALTHY DONOR → PBMC COLLECTION
Start with a healthy donor
Every batch begins with T-cells collected from a single healthy donor. One donation creates therapy for many people.
ALLOGENEIC · ONE SOURCE, MANY PATIENTS
Step 02 / Engineer
PURIFY · EDIT · ARM · EXPAND
Sofi-cel:
Engineer the cells
Purified donor T-cells are activated, then CRISPR-edited to remove CD7 and the TCR. A lentivirus adds the CD7-targeted CAR, and the new sofi-cel multiplies into many doses.
ACTIVATE · CD7/TCR KO · ANTI-CD7 CAR · EXPANSION
Step 03 / Cryostore
HARVEST · FILL · CRYOPRESERVE
Freeze into many doses
The expanded cells are filled into vials and cryopreserved. One donor creates material to treat many patients, ready off-the-shelf.
CRYOPRESERVED · QC-RELEASED · STORED
Step 04 / Infuse
THAW · INFUSE
Ready when the patient needs it
When a patient needs treatment, a vial is thawed and infused. The manufacturing process enables immediate, on-demand use
SAME-DAY READY · NO MANUFACTURING DELAY
Step 05 / Strike
CIRCULATE · DOCK · LYSE
Seek and destroy
The sofi-cel circulates through the blood until they meet a pathogenic cell carrying CD7. The CAR locks onto the pathogenic cell, eliminates it and then moves to the next, and the next…
SURVEILLANCE · SERIAL CYTOTOXICITY · CD7+ MALIGNANCY
Step 06 / Recover
CLEARANCE · THE GOAL
Eliminate the cancer cells
Cell by cell, the therapy helps clear the disease, giving the patient a path back to health, from one healthy donor’s gift.
