One donor.
A hundred doses.
Ready when the patient is.
How Sofi-cel, Allotera’s allogeneic, gene-edited CAR-T therapy, is built from a single healthy donor and put on the shelf, ready to fight T-cell cancers.
The Off-the-Shelf Journey · 9 Steps
Step 01 / Source
HEALTHY DONOR → PBMC COLLECTION
Start with a healthy donor
Every batch begins with T-cells collected from a single healthy donor, never the patient. One donation seeds therapy for many people to come.
ALLOGENEIC · ONE SOURCE, MANY PATIENTS
Step 02 / Culture
ISOLATE · ACTIVATE · EXPAND
Purify, activate & culture
The T-cells are purified from the donor’s blood, switched on with activation signals, and cultured so they begin dividing, building the population we’ll engineer.
T-CELL ISOLATION · ACTIVATION · EXPANSION PRIMED
Step 03 / Edit
CRISPR/Cas9 · ELECTROPORATION
Remove CD7 and the TCR
A pulse of electroporation delivers CRISPR/Cas9 to knock out CD7, so the cells won’t kill each other, and the TCR, so they won’t attack the patient.
CD7 KO → NO FRATRICIDE · TCR KO → NO GvHD
Step 04 / Arm
TRANSDUCE · ANTI-CD7 CAR
Add the CD7-targeted CAR
We arm each cell with a chimeric antigen receptor aimed at CD7. The edited donor T-cell is now Sofi-cel, a purpose-built cancer hunter.
RECEPTOR + COSTIM + CD3ζ · SOFI-CEL
Step 05 / Expand
1 → HUNDREDS OF MILLIONS
Grow the army
The engineered cells multiply again and again. One becomes hundreds of millions, far more than a single patient needs.
MASSIVE EXPANSION · MULTI-DOSE YIELD
Step 06 / Cryostore
HARVEST · FILL · CRYOPRESERVE
Freeze into 100+ doses
The harvest is filled into more than a hundred vials and cryopreserved. Finished doses sit ready on the shelf, no patient-specific batch required.
CRYOPRESERVED · QC-RELEASED · STORED
Step 07 / Infuse
THAW · INFUSE
Ready when the patient is
When a patient needs treatment, a vial is thawed and infused. There’s no weeks-long wait to manufacture a one-off, personalized batch.
SAME-DAY READY · NO MANUFACTURING DELAY
Step 08 / Strike
CIRCULATE · DOCK · LYSE
Hunt and destroy
The Sofi-cel cells circulate through the blood until they meet a cancer cell carrying CD7. The CAR locks on and kills it, then releases and moves to the next, and the next.
SURVEILLANCE · SERIAL CYTOTOXICITY · CD7+ MALIGNANCY
Step 09 / Recover
CLEARANCE · THE GOAL
Clear the cancer
Cell by cell, the therapy clears the disease, giving the patient a path back to health, from one healthy donor’s gift.
